Xencor's lead protein therapeutic, XPro™1595, is in pre-clinical development for treatment of rheumatoid arthritis, a chronic degenerative disorder. XPro™1595 is a first-in-class Dominant Negative inhibitor of TNFα (DN-TNF) that we have designed to act through a new molecular mechanism of action. The inhibition of TNFα has been clinically proven to be an effective treatment of rheumatoid arthritis and other inflammatory diseases. Annual sales of TNFα blockers currently exceed $8.5 billion.
XPro™1595 was designed using our Protein Design Automation® technology to function as a ligand and target receptor-specific inhibitor of TNFα, and thereby provide a greater safety margin and unique efficacy profile when compared to existing TNF inhibitor drugs. XPro™1595 demonstrates potent activity in animal models of rheumatoid arthritis while not suppressing the immune response to infection, a side effect of current biologics therapies against TNF.
XPro™1595 also has been engineered for superior pharmaceutical properties, such as efficient microbial manufacturing, high stability and a convenient dosing schedule. XPro™1595 is being developed for the treatment of rheumatoid arthritis and other inflammatory diseases and will be administered as a subcutaneous injection.
Xencor has engineered Dominant Negative inhibitors of other members of the TNF structural superfamily such as RANKL for the treatment of metastatic bone disease and osteoporosis, and BLyS/BAFF for lupus and other autoimmune diseases.
